Does the (new) ETC Puck Base (p/n: 4380A11430), which looks like it's based on the ASRock NUK Box, support DisplayPort via the front facing USB-C ports?

No, the front USB-C port cannot be used for DisplayPort. Only the external display ports on the back are supported, one DisplayPort and one HDMI.
